General Experience:

             Bacteria isolation, purification and identification from different environmental samples

             Identification of cultured and uncultured bacteria

             DNA isolation from gram negative and gram positive bacteria and also from soil

             PCR amplification using universal or specific primers, cloning, Gene libraries, Screening of the gene libraries,    

               sequencing, sequence analysis,

             The use of SSCP-PCR and others PCR-based approaches to recognize between bacterial species at the strain

               level.

             Probes and primers design for the functional genes

             Screening and Constructing of genetically modified bacterial degraders  for the Aromatic compounds e.g.  Dioxin

              and PCBs

             Phylogenetic analysis of 16S ribosomal RNA genes either for cultured or yet uncultured microbial species from

               different environment Ecosystems

             Amplification of specific fragments of nucleic acids (DNA, RNA) using   polymerase chain reaction (PCR)

             Detection and separation of PCR products using agarose gel electrophoresis.

             Phage and phagmid library

             Fosmid library

             Overexpression of genes 

             Protein electrophoresis

             Isolation of the function fraction protein by FBLC

             Identification of the metabolites by HPLC, HPLC-Mass spectrum, and NMR

             Cloninig in gram negative and gram positive bacteria by using vector and also shuttle   vector

             Biodegradation of aromatic compounds and its application in bio-electrochemical systems

             Genetic characterization of antibiotic resistance and virulence factors in bacteria

 

 

Projects:

 

• Principle Investigator: Methods for screening dioxygenase enzymes and organisms that will catalyze the hydroxylation of a variety of aromatic substrates, and using the hydroxylated products in the applications for medicine in the pharmaceuticals manufacturing (STDF) (2009) (successfully finished 2014 )

 

• Principle Investigator: Plastidic expression of X- or N-containing xenobiotic aromatic compounds degradative dioxygenase for the bioremediation and production of pharmaceuticals compounds
